Drug Discovery Short-Circuits Cancer Growth

A new drug that blocks cancer's main source of growth has been created in the lab and proven effective in mice, scientists are reporting. It is now being readied for clinical trials in patients.

Far more potent than similar compounds already in clinical trial, the drug short-circuits the normal ability of cells to sense the need to grow and divide -- a signal that cancer cells exploit to spread in the body.

The scientists are working with clinicians to test the drug's effectiveness against a range of cancers that have proven difficult to treat.

The discovery is reported in the Feb. 10, 2009 edition of "PLoS Biology," a journal published by the Public Library of Science.

The research was led by scientists at the University of California, San Francisco (UCSF) and the UCSF Helen Diller Family Comprehensive Cancer Center (http://cancer. ucsf. edu/). Senior author of the paper is Kevan Shokat, PhD, Howard Hughes Medical Investigator and professor of cellular and molecular pharmacology at UCSF.

Normally, in response to growth signals, a multi-protein unit in cells called mTOR integrates information about the cell's nutritional and energy needs, and prompts the cell to manufacture key proteins for cell growth. But cancer exploits this signal for its own growth.

Clinical trials are underway to stymie cancer proliferation by using a drug called rapamycin--marketed as Rapamune--or related compounds to block the growth signal cycle. The new drug greatly improves on rapamycin's effectiveness, the scientists reported. The name mTOR stands for "mammalian target of rapamycin."

Of serious concern to clinicians, rapamycin and related drugs can actually promote cancer at the same time they thwart it. This happens, the scientists found, because the drugs only partially block the cells response to a growth signal. When this happens, the drugs end up augmenting the growth signal itself because a feedback process in the cell kicks in to assure adequate nutrition. With the feedback system in play, cancer cells can regain needed nutrients and continue to proliferate.

The new drug totally blocks this feedback loop, said Shokat, who also is a faculty affiliate at the California Institute for Quantitative Biosciences, known as QB3, which is headquartered at UCSF.

"We were trying to synthesize compounds that could help us learn more about how cancer exploits normal growth controls," he explained. "Once we made it, though, we found it was even better than we thought it would be at blocking mTor signaling. It does everything that rapamycin does and more."

The new drug succeeds because there are two mTOR signal pathways, and it blocks both, the scientists found. Rapamycin only blocks one, and so allows the growth-signaling system to still function.

The scientists think that the drug's total blockage of the nutrient-sensitive mTOR and its feedback loop offer a major advance over rapamycin based drugs, which have been approved to treat only renal call carcinoma effectively.

"I hope the new drug can be used to treat a range of cancers," Shokat said. "We will work with clinicians to test it against a number of types of cancer - colorectal, lung, breast, multiple myeloma and others. We want to first find the cancer that is most sensitive to it."

The new compound has been dubbed a TORKinib because it inhibits the mTOR signal. UCSF has applied for a patent and licensed the patent to a startup biotech company, co-founded by Shokat and colleagues, to advance its use in clinical trials to treat cancer.

MTOR is known as a kinase, a ubiquitous type of signaling molecule - there are more than 500 different kinases in the body - that essentially switches proteins on or off. The switch is one of the most common interactions in the body. The kinase adds a small molecule known as a phosphate group to the protein, and that single action either turns on the protein or dampens its activity.

Like all signaling systems in the body, the one involving mTOR goes through many steps to accomplish its duties. The different steps form a cascade that can ramp the signal up or down, depending on the conditions of the cells or tissues. These kinase cascades are embedded within complicated feedback loops, such as the one activated by rapamycin.

Research on the mTOR pathway is of increasing interest to drug companies, Shokat said.

Since mTOR acts "upstream" and "downstream" of other key kinases that are found to cause cancer-- such as the much-studied kinases PI3K and Akt-- he thinks blocking it will short-circuit the many feedback loops cancer cells use to generate and maintain a growth signal.

"We are extremely excited about the potential of targeting mTOR in this way to treat a number of cancers, although we are aware that there are many hurdles to reaching the finish line," Shokat said.

Lead author on the paper is Morris E. Feldman, a graduate student in Shokat's lab.

Shokat's colleague, Davide Ruggero, PhD, who studies cancer's effect on protein synthesis, is also a coauthor. Ruggero is assistant professor of urology in the UCSF Helen Family Comprehensive Cancer Center.

Other coauthors are Beth Apsel, PhD, a graduate student in chemistry and chemical biology; and Zachary Knight, PhD, a postdoctoral scientist in Shokat's lab during the research project.

The research was funded by the Howard Hughes Medical Institute.

The private company co-founded by Shokat is called Intellikine. He is the chairman of its scientific advisory board and holds stock options in the company.

Los Angeles, California (PRWEB) – Jeannette Medelus is a 45-year old woman who can barely see to do her daily chores. Etienne Salomon is a 67-year old man who has to be led around by his grandchildren because he cannot see. The nearest medical eye facility is in Port-au-Prince; and even if they could get there, they could not afford to pay for the treatment. They both know that if they go blind, they might as well die. These are just two of nearly 700 people anticipating the arrival of the Medical Team to have their eyes examined by the physicians provided by Friends of the Children of Lascahobas, Haiti. Both patients are seen and successfully treated. Jeannette only needed eyeglasses, but Etienne had to have cataract surgery. They both leave praising the physicians and stating in French, “God will save a place in Heaven for you.”

Friends of the Children of Lascahobas, Haiti (FCLH) is a non-profit volunteer organization dedicated to providing much needed services to the villagers of Lascahobas, an isolated and impoverished mountain village. Besides providing the yearly medical humanitarian mission, FCLH educates and instructs women in small business management skills, feeds 300 children in their Nutrition Program, and staffs and equips the Hospital of Lascahobas that FCLH built and maintains.

FDG Fine Art Gallery, Los Angeles Urban Business Journal, Underground Frames, and the Westfield Eye Center (located in Las Vegas, Nevada) will sponsor a Haitian Art Exhibit and Reception to benefit Friends of the Children of Lascahobas, Haiti on Saturday, August 2, 2003, from 6:00pm – 10:00pm, at the FDG Fine Art Gallery, 4470 West Adams Boulevard, Los Angeles, California. The exhibit features original Haitian oil paintings, sequined flags, masks, and folk art presenting work from over 20 different artists.

Dr. Kenneth Westfield, FCLHÂ’s chief of medical operations, recruited local organizations, friends, and relatives to assist with fundraising efforts to benefit FCLHÂ’s initiatives.

Many people are stunned by the heartbreaking stories told by Dr. Westfield of poverty and lack of medical care. “It’s amazing. On the first day the medical team arrives to work at the hospital, we are greeted by hundreds of people. The people will camp outside the hospital to wait their turn to see the physicians,” says Dr. Westfield. “As soon as we unload and set up the equipment, we prioritize their conditions and immediately begin treatment. We will examine all the people, perform the surgeries and be ready to return to the United States within four days.”

FCLH founded in 1976 by Estelle Dubuisson, a Lascahobas native and retired Brooklyn medical technician. Dr. Westfield states, “By educating the people, feeding the children and preserving the health of many townspeople, her efforts have helped to reduce the villagers’ suffering.”

Although progress has been made, the needs are tremendous. Dr. Westfield said, “The hospital is staffed with only one general practitioner for 56,500 people, and needs additional staff, medical supplies and equipment. Since September 11, 2001, it has become increasingly harder to get supplies and equipment to Haiti. Green Mountain Coffee Survey Gauges Bostonians’ Coffee Habits And Fair-Trade Savvy

A survey conducted on behalf of Green Mountain Coffee® shows what many of us would already suspect: Boston is a coffee-crazed town. In fact, according to Green Mountain Coffee’s Fair Trade-Out Survey, 1 in 3 Bostonians confess that they believe they need coffee every day, and on average drink 2.4 cups of coffee per day. Almost 1 in 5 (17%) drinks four or more cups daily.

But even with all the coffee drinking in Beantown it seems very few in the Hub know much about the coffee in their cup, including how to make it, where it comes from, and perhaps most importantly, how their choice of coffee can have an impact on communities around the world. Conducted in celebration of Fair Trade Month in October, The Fair Trade-Out Survey explores Boston’s relationship with coffee and aims to educate coffee drinkers on the benefits of trading out their coffee for Fair Trade Certified™ varieties. Fair Trade provides a fair, minimum price to coffee farmers for their beans, resulting in higher quality coffee, and a higher quality of life in coffee-farming communities.

What would Bostonians Trade Out for Their Coffee?
When faced with the choice of losing their daily cup of coffee, Boston-area coffee drinkers would trade out most anything to keep it, including their car (23%), mobile phone (52%), and their pet (34%). What else would Bostonians trade out to keep their coffee?
 About the same percentage of Bostonians would “absolutely” rat out Whitey Bulger (28%) as would talk Tom Brady into cutting his new flowing locks of hair (26%).  Just as many Boston-area residents (1 in 3) would opt to pick up someone at Logan Airport every week for a month as would party with Deval Patrick.  And when it comes to bravery, almost the same number of Bostonians (44%) would keep open the option of either fighting Bruins’ would-be pugilist Shawn Thornton or swimming across the Charles River before giving up their coffee. Overall, Boston men would go to greater lengths than women to keep their coffee:
 So much for man’s best friend! Nearly twice as many men (41%) as women (25%) would trade out their pet before giving up their coffee.  Baby, You Can Drive My Car: More men (25%) than women (19%) would trade out their car to keep their coffee.  What would women do?: The one thing women would do more than men when it comes to keeping their coffee is volunteer at a non-profit for a month, with more than half of women (56%) willing to volunteer vs. 1 in 3 men (34%). Conducted shortly after Boston became a Fair Trade Town in September, The Fair Trade-Out Survey is part of a campaign to encourage Boston-area residents to trade out their usual cup of coffee for Green Mountain Coffee’s Fair Trade Certified blends.

“The Fair Trade-Out Survey is a fun way to remind people that coffee is more than just a part of a daily routine,” said Sandy Yusen, Director of Public Relations, Specialty Coffee Business Unit of Green Mountain Coffee Roasters, Inc. “The choices we make about what’s in our cup have the power to improve the lives of coffee farmers and their families all over the world.”

The Fair Trade-Out Survey underscores the opportunity to promote better understanding of the benefits of Fair Trade. Currently, only 1 in 8 (13%) Bostonians regularly buy Fair Trade Certified products – that’s just one-third of those who regularly buy local products (38%). The survey suggests that this may be due to lack of education on what Fair Trade means:

 2 in 3 (68%) Bostonians stated that they did not fully understand what Fair Trade means, and 1 in 3 (30%) admitted they had no idea. Even when given options, less than half (48%) could identify the definition of Fair Trade.  However, after reading an explanation of the meaning of Fair Trade, 3 in 4 (76%) Bostonians were likely to purchase Fair Trade Certified products in the future.  When it comes to paying a premium for Fair Trade products, 26% of those surveyed agreed that they would be willing to pay 5% to 10% more.

Boston’s Coffee Culture
When it comes to a “regular” coffee, Bostonians have a much different view than the rest of the country. In Boston, a “regular” most often contains cream and one or two sugars (48%), where nationally 57% feel a “regular” is served black.
 There’s no such thing as an occasional coffee drinker: 78% of Boston-area respondents drink coffee every day. Only 1 in 10 (10%) drinks coffee less than 5 days per week.  And when Bostonians indulge, they really indulge: only 1 in 4 respondents (27%) stop at one cup of coffee per day. In fact, 1 in 5 (17%) drinks four or more cups daily!  Make mine black: 27% of respondents nationally take their coffee black, while only 18% of Bostonians enjoy the brew in its purest form. Men are more likely to take their coffee black than women (22% vs. 14%).  Half and half is the most popular coffee lightener: One in four (23%) Bostonians prefer half & half over flavored creamer (13%), and whole milk and cream (both 11%).  Most popular place to drink coffee? An overwhelming 76% drink coffee at home, followed by the office at 12%.  On average, men drink more coffee than women. Boston men drink 2.4 cups per day; women drink 2.2 cups per day. When it comes to heavy coffee drinkers, only 1 in 8 women (13%) surveyed drink 4 or more cups per day, compared to 1 in 5 men (21%) who fill up 4 or more times throughout the day.  Bostonians are sweeter: More than half of Boston respondents (67%) use sugar or sweetener.

Unlimited server and personal certificates at one fixed fee; New service offered, in partnership with Comodo, as part of expanding InCommon Trust Service Portfolio

The InCommon Federation has today announced the launch of a new trust service that will offer unlimited server and personal certificates to U. S. colleges and universities. As the first certificate service of its kind in the U. S., Internet2, which operates InCommon, is introducing this offering to provide the U. S. higher education community with a cost-effective means of deploying and operating a more secure and authenticated online environment for their campus constituencies.

“This new service provides tangible value to the U. S. research and education community and extends a pathway for innovation on campus,” said John Krienke, chief operating officer of InCommon. “By offering unlimited SSL certificates, we’re providing the potential for campuses to significantly reduce their certificate-related expenses. But at least as important, later this year we will include unlimited personal certificates, setting the stage for campuses to more easily adopt important services like signed email and secondfactor authentication. Economics are no longer a limitation for certificate possibilities."

InCommon is offering this service through a partnership with Comodo CA Ltd., a major certification authority and Internet2 industry member. Through the agreement, U. S. institutions of higher education can purchase unlimited numbers of certificates for SSL, personal signing, encryption, and code signing. InCommon partnered with Comodo, in part, because of the successful implementation of a similar program the company developed with TERENA (the Trans-European Research and Education Networking Association). The program launched last year has enabled its NREN members in over 30 European countries and international public sector organizations to offer certificates powered by Comodo to all education institutions throughout Europe.

“We are excited about the opportunity to join InCommon in helping millions of students in colleges and universities across the U. S. to secure their online experience and communications,” said Melih Abdulhayoglu, chief executive officer of Comodo. “Enabling students and faculty members with the personal secure certificate and second factor authentication, whether for email encryption or for secure access to the university’s internal network, will prevent security breaches and leak of digital information.”

Any higher education institution with its primary location in the U. S., and qualifying for an “edu” domain name, is eligible for the program. Internet2 member institutions will receive a 25 percent discount on the Certificate Service. Institutions must also participate in or join InCommon to take advantage of the Certificate Service.

The service fee schedule is based on an institution’s classification under the Carnegie Foundation for the Advancement of Teaching and requires an initial three-year commitment.

The InCommon Certificate Service is testing the program with a small number of institutions during June 2010. A full rollout of the service is expected in late summer of 2010. Institutions with expiring certificate agreements will receive priority, allowing them to avoid service interruptions. The roots of InCommon's SSL and personal Certificate Authorities are in all of the major Internet web browsers and devices.

Coker College is pleased to announce the hiring of Tony Smith as the Cobras’ first ever men’s lacrosse coach.

Coker College is pleased to announce the hiring of Tony Smith as the Cobras’ first ever men’s lacrosse coach. Smith will take the helm of the newly formed Coker lacrosse program starting December 1.

“I would like to thank Coker College Athletic Director Lynn Griffin and Coker College President Dr. Robert Wyatt for the opportunity to be the inaugural head coach of the men's lacrosse team,” said Smith. “I look forward to building the Coker College men’s lacrosse program into a Conference Carolinas and NCAA regional powerhouse.”

Smith comes to Coker from fellow Conference Carolinas member Belmont Abbey College, where he has been the assistant coach and defensive coordinator since 2007. At the Abbey, Smith was in charge of all of the Crusaders’ recruiting efforts locally, nationally and internationally, while also creating and implementing the defensive unit and strategy for a lacrosse program that has finished in the top three in the Conference Carolinas standings in two of the last three years.

In 2010, the Crusaders defensive unit held opposing teams to less than 10 points per game. The 2010 season was also the second consecutive season that the Crusader defensive unit caused more than 100 turnovers, with 126.

In addition to his coaching responsibilities, Smith was also an instructor in Belmont Abbey’s Sports Administration Program.

“I would like to thank Coach Jim Dietsch and Dr. Travis Feezell, both of Belmont Abbey College, for being invaluable in my development as a college coach and college professional,” Smith said. “I especially want to thank Coach Dietsch. Jim has been my mentor and friend for almost twenty years. One could not find a better lacrosse and "life lessons" mentor than Jim Dietsch. I owe all of my coaching acumen to Jim and I will be forever grateful.”

A graduate of the University of Maryland-College Park, Smith holds bachelor’s degrees in history, as well as government and politics. He earned his master’s of education in sports administration from Wingate University, and prior to his time at Belmont Abbey, Smith served as an assistant coach at Wingate University and the Community College of Baltimore College-Catonsville.

Smith played collegiately at the Community College of Baltimore College-Catonsville (1994-1996) and Whittier College (1996-1998).

“I am honored that Tony Smith will become the inaugural men’s lacrosse coach at Coker,” said Griffin. “He has the vision to develop a competitive lacrosse program that Coker College will be proud of. I feel that his experience at the NCAA Division II level will aid him in recruiting the type of student-athlete that can excel here.”

“I am looking forward to beginning to aggressively recruit potential student athletes to fill a complete roster by the start the fall 2011 academic semester,” said Smith. “This recruiting will begin immediately. My hope will be to instill sound liberal arts academic values, moral integrity and a "win now" attitude in my future team as we begin to build a successful lacrosse program here at Coker.”

Coach Smith embodies all of the characteristics we believe define a successful college coach,” said Wyatt. “His focus is on recruiting student-athletes who are concerned about their performance in the classroom, as well on the field, along with his tremendous playing and coaching experience will help establish a winning tradition for Coker lacrosse.”

Coker becomes the seventh Conference Carolinas member to add lacrosse. Conference Carolinas is known as one of the top conference for lacrosse in the country with many highly successful programs such as two-time NCAA Division II National Champion Limestone College.

The 14th NCAA Division II athletic program at Coker, the Cobra lacrosse program will begin in the fall of 2011. They will practice and play games on the football field at Byerly Park in Hartsville.

On May 10, Alan Gathright of the San Francisco Chronicle reported on a regional human rights organizationÂ’s demand that San Jose, Calif. Police halt stun gun use immediately. The Coalition for Justice and Accountability cited concerns that other groups have over the past months.

“We have heard the human rights organizations’ concerns time and time again,” said Siciliano. “While nobody is ever 100 percent wrong, these people come pretty close to it when they talk about stun guns.”

"Yes, stun guns come with risks," Siciliano continued. "No weapon is 100 percent safe, but this has never been the point.”

“If police opt to use stun guns more often than their firearms,” Siciliano added, “the net gain will be much safer circumstances for the public. This will be especially true if we develop strict guidelines for usage, as some police departments and associations of law enforcement professionals already have."

The Kansas City Star’s Christine Vendel reported on May 10 that Kansas City police have “adopted a policy [that] clarifies situations in which officers can use” stun guns.

The May 8 editorial page of Florida Today, in a piece calling for more restrictions on stun gun use, cited Brevard County, Fl. police chiefs who have also recommended new guidelines for stun gun use.

“Industry and law enforcement are moving in the right direction on stun gun use,” said Siciliano. “Soon, the extremists’ exaggerations will be evident. We must reign in the hyperbole and think clearly about stun guns, which are here to stay.”

The Associated Press has reported on the rise of Stinger Systems, a North Carolina-based company that has shipped samples of its stun guns to more than 1,000 interested law enforcement agencies. Bob Gruder continues in his roles as chairman and chief executive at that company.

Another stun gun manufacturer is North Carolina-based Law Enforcement Associates Corporation (LENF). A late March press release from LENF announced favorable safety results from an International Electrotechnical Commission test of the firm's stun gun.

A group of teenagers starts a project that will sharpen emergency response by making homes easier to locate.

The city of Statesville is under the lens this week with the start of a new project by a local teen. Andy Laws, age 16, has a plan that would put Statesville on the technological map -- literally. His idea entails gathering photos of houses in the city and creating an online database.

Housephotos. org is a free application that provides photographs of homes by street address. Laws was inspired to begin the project in hopes that emergency services, such as first responders, would utilize the database for more effective responses. The site is dynamic in the fact that it has many different uses. Domino's Pizza was contacted by e-mail and showed genuine interest in the project for delivery purposes.

Knowing the physical appearance of a home will also make it easier to locate, which has raised a few questions from the community. Various security measures are in place to insure the integrity of the database, and to keep the public as safe as possible. Homeowners can choose to contact the site by e-mail if they want their listing removed, though Laws discourages it and with good reason.

The teenage volunteers of the site are referred to as the Street Team. The group is composed of various teenagers from around Iredell County, who see the site as a way to positively affect the community. The crew hopes to finish the project by 2009, a time when they will have to disband to further their education after high school.   Not getting a semen analysis right away. It is estimated that 20-30 percent of infertility (http://www. ivf1.com) cases are due to the male factor.   Over-diagnosing an ovulation, a condition in which the ovary does not release a ripened egg each month as part of a woman's normal cycle. Many doctors will diagnose an ovulation by using a blood test for a hormone called progesterone. Progesterone is only elevated in women after they have ovulated. An elevated level of progesterone that is consistent with ovulation is anything over 2.5 or 3.0. However, many doctors will mistakenly tell a woman she has not ovulated if her progesterone level is less than 10. "This is a very common mistake that results in women being misdiagnosed, having unnecessary tests performed, and being treated incorrectly," says Morris.   Many doctors order blood work at the wrong time of a woman's cycle. Hormone levels in women change according to the time of the menstrual cycle. Every test needs to be timed precisely. Failure to recognize this fact results in hormone testing being performed with un-interpretable results. For example, the post coital test looks for sperm in a woman's cervix after a couple has had intercourse. In the past, it was believed that failure to find moving sperm could be a sign of a woman "killing off" her partners' sperm. However, several rigorously performed studies have shown that the test is worthless. However, state grants and scholarships offered through the private sector are available to help pay increasing tuition cost. A slow economy has caused a sharp drop in college and university state spending. According to The National Center for Public Policy and Higher Education, public four-year college tuition in Ohio has increased 17%. The result - a higher cost for higher education

4. Local Organizations - There is a better chance of winning money from local organizations such as churches, clubs, community groups, and unions since fewer students are likely to apply. 5. Place of Employment-Employers may also offer grants and scholarships. Inquire at your personnel office. Dependent students should ask their parent or legal guardian to check the availability of awards.

6. Announcements - Keep your eyes open. Take time to read bulletin boards, posters, and articles in newspapers for competition announcements. Some scholarships are episodic and may occur only once.
